A Metal Organic Framework-Based Light Scattering ELISA for the Detection of Staphylococcal Enterotoxin B

Enzyme-linked immunosorbent assay (ELISA) is one of the most commonly used method for detection staphylococcal enterotoxin B (SEB), main protein toxin causing food poisoning. However, traditional ELISA reaction needs to be stopped by sulfuric acid obtain stable colorimetric signal, and it easily influenced a colored sample. In order address this problem, new using zeolite imidazolate skeleton-8 metal-organic framework (ZIF-8 MOF) as light scattering (LS) reporter SEB was developed in work. ZIF-8 MOF has characteristics high porosity, large specific surface area, clear pore structure, adjustable size, which representative MOFs constructed from Zn2+ 2-methylimidazole (2-mIM). The 2-mIM ligand exhibited antioxidant activity can strongly react with H2O2, could destroy structure ZIF-8, resulting obvious decrease LS intensity. We combined sandwich immune construct successful SEB. This more reliable than commercial tests kits samples, simple, sensitive, selective, great potential other toxins simply changing corresponding recognition units.
